臍帶是哺乳類的連接胎兒和胎盤的管狀結(jié)構(gòu),由兩條動脈和一條靜脈構(gòu)成。臍帶呈圓柱狀結(jié)構(gòu),內(nèi)有兩條臍動脈及一條臍靜脈。臍帶內(nèi)有一條臍靜脈,接受從母體來的新鮮血液并流經(jīng)胎兒全身回到胎兒的動脈系統(tǒng),最后進(jìn)入兩條臍動脈流入胎盤至母體,這是胎兒血循環(huán)的特點之一。胎兒藉以生存的營養(yǎng)物質(zhì)如電解質(zhì)、無機(jī)鹽、激素、微量元素、免疫抗體等,均需經(jīng)過臍帶傳送。

實驗材料和器具:
樣品:臍帶組織
工具:凈信全自動樣品快速研磨儀
耗材:專用研磨罐,液氮、剪刀、鑷子等
實驗步驟:
1、準(zhǔn)備研磨罐和樣品;
2、取組織剪成小塊放入研磨罐中,將蓋子蓋緊;
3、將研磨罐置于裝有液氮的容器內(nèi),液氮容量沒過研磨罐冷凍幾分鐘,取出時戴手套或使用鑷子夾取避免液氮凍傷;
4、首先將適配器放入研磨儀中固定,然后在將研磨罐放置適合器內(nèi),以保證研磨時的受力平衡,旋緊垂直定位螺旋,將安全鎖扣緊。
5、設(shè)置參數(shù)啟動程序結(jié)束后即可得到破碎粉末;
6、研磨完畢后請關(guān)閉電源,將安全鎖歸位;
7、完成后續(xù)RNA提取步驟。
